Our Core is a collaborative effort.
- Pluripotent Stem Cell Reprogramming, Genome Editing, and the Cell Bank are currently located at the UConn Health campus in Farmington.
- The Genetics and Genomics Division, including the Chromosome Core, is located at UConn Storrs campus.
- Single Cell Genomics is based at the Jackson Laboratory for Genomic Medicine in Farmington.
Services
Reprogramming Services
We provide reprogramming service for the following tissue types:
- Dermal Fibroblasts – as live cell culture or cryopreserved in vial(s)
- Peripheral Blood
- Umbilical Cord Blood – frozen
If you would like to discuss the possibility of reprogramming other unique cell types, please contact us.
Cell Lines
Distribution of Human Pluripotent Stem Cell Lines
-hESCs from UConn Stem Cell Core: The Uconn Stem Cell Core has generated four hESC lines (CT1, CT2, CT3, and CT4) that are NIH Human ESC Registry-approved and available to researchers.
-hiPSCs from UConn Stem Cell Core. The Uconn Stem Cell Core has generated several control hiPSC lines from a variety of source cell types (fibroblasts, umbilical cord blood, and peripheral blood) and using multiple reprogramming techniques (retroviral, STEMCCA, mRNA, episomal, and Sendai virus). These lines are available to researchers upon request.
-IPSC Rare Disease Models. The Uconn Stem Cell Core has collaborated with the Foundation For Prader-Willi Research to establish a Biobank housing high-quality iPSCs derived from individuals with Prader-Willi syndrome. These iPSC lines have undergone various quality control measures and are available upon request to both academic and industry researchers.
Genome Editing: Custom TALEN Design
Services include:
- Generation of TALEN and sequencing
- Verification of TALEN
- In-Vitro or In-Vivo expression on exongenous DNA
- In-Vivo expression on genomic locus
Genome Editing: Custom CRISPR Design
Services include:
- CRISPR construction and sequencing
- Validation
- In-Vitro or In-Vivo on exogenous DNA
- On genomic loci
Genome Editing: Vectors for Targeting
Services include:
- Generation of targeting vector
- Endo-free DNA prep for targeting
- Linearization of targeting vector
- Design and ordering of single stranded ligo for targeting vector
Genome Editing: Cell Culture Services
This service can be used to perform cell targeting experiments, selection and genotyping of resistant clones, expansion and freeze of positive cells clones. Cells are targeted by electroporation and plated to perform selection with antibiotics. Number of clones from any targeting experiment can vary widely so fee structure is based upon a per clone basis.
Other Services
Validation and Quality Control Analysis
- Verification of Pluripotency
- Mycoplasma Testing
Cell Biobanking: We will perform QC measures, expand, store, and distribute your custom hESC and iPSC lines when requested by your collaborators. We can also provide continuous banking as a back-up repository for your lab.
Training: We provide training in iPSC/hESC Basic Culture Techniques, consulting, and troubleshooting support. Individual workshops are available for iPSC/hESC differentiation to neural, cardiomyocyte, and vascular endothelial lineages.
